Reference

Preparation of Rothera's power
  1. Weight following chemical & mix thoroughly
  2. Sodium carbonate ­­­­­­­= 20 gm
  3. Ammonium sulfate ­­­­= 20 gm
  4. Sodium nitroprusside ­­­= 1 gm (make fine powder of it with mortar and pastle before mixing)
Sample
Procedure
  1. Take pinch of powder on a glass surface or a tile or a tile with well in it
  2. Put a drop of sample on powder
  3. Do not take sample more than what powder can absorb.
Observation and interpretation
  1. If purple color observed, ketones are reported as “positive”
  2. If no purple color observed, ketones are reported as “negative”
  3. Read IQC (as shown below) to prevent confusion in detection of color
Internal Quality Control
  1. Frequency of IQC : Whenever sample is received, Both the control (Positive & negative ) has to be run along with test.
  2. Prepare 20 ml urine pool from OPD samples of apparently healthy individual. Divide in to two parts.
  3. Preparation of Positive control
  4. In 10 ml of pooled urine- add 10 ul acetone.
  5. Prepare aliquotes of 200 ul and give lot number e.g KP1, KP2, KP3….
  6. -Preparation of Negative control
  7. Take 10 ml of pooled urine
  8. Prepare aliquotes of 200 ul and give lot number e.g KN1, KN2, KN3….
  1. Take powder in two wells.
  2. In one well add negative control.
  3. In another well add positive control.
  4. Note observation in Ketones IQC Sheet
  5. Upload sheet every month in LISPrint sheet
  6. The monthly QC sheet has to be uploaded in KTO calibration file.
External Quality Control

Currently there is external quality Control done in the laboratory.
