Reference

Preparation of Rothera's power
  1. Weight following chemical & mix thoroughly
  2. Sodium carbonate ­­­­­­­= 20 gm
  3. Ammonium sulfate ­­­­= 20 gm
  4. Sodium nitroprusside ­­­= 1 gm (make fine powder of it with mortar and pastle before mixing)
Sample
Procedure
  1. Take pinch of powder on a glass surface or a tile or a tile with well in it
  2. Put a drop of sample on powder
  3. Do not take sample more than what powder can absorb.
Observation and interpretation
  1. If purple color observed, ketones are reported as “positive”
  2. If no purple color observed, ketones are reported as “negative”
  3. Read IQC (as shown below) to prevent confusion in detection of color
Internal Quality Control
  1. Frequency of IQC : Whenever sample is received, Both the control (positive and Negative) has to be run along with test.

Preparation of positive control

  1. Prepare pooled serum of 10 ml from OPD sample
  2. Add 10ul Acetone in 10 ml of pooled serum
  3. Give lot number as KP001, KP002,KP003…etc
  4. Fill 50ul in each ependroff cup and seal it with parafilm and labeled as KP001, KP002,KP003…etc
  5. Store ependroff cups in refrigerator at 2-8 ' C.

Preparation of Negative control

  1. Prepare pooled serum of 10 ml from OPD sample
  2. Give lot number as KN001, KN002,KN003…etc
  3. Fill 50ul in each ependroff cup and seal it with parafilm and labeled as KN001, KN002,KN003…etc
  4. Store ependroff cups in refrigerator at 2-8 ' C.

How to do Quality control

  1. Take powder in two wells.
  2. In one well add Positive control
  3. In another well add Negative control.
  4. Note observation in Ketones IQC Sheet
  5. Upload sheet every month in LIS(Docs_Qualitative IQC)ketone_sheet.ods
External Quality Control

QA Forum sample for Ketone bodies done every 6 monthly.
