SOP FOR DECALCIFICATION

1. PURPOSE OF EXAMINATION: lay down the procedure for BONE DECALCIFICATION in Histo-pathology section NCHLS Surat.

2. PRINCIPLE AND METHOD OF PROCEDURE USED FOR EXAMINATION: In order to obtain satisfactory paraffin section of bone, inorganic calcium must be removed from the organic collagen matrix calcified cartilage and surrounding tissues by adding acid to form soluble calcium salts.

3. STAFF RESPONSIBLE: Resident doctors under supervision of tutor.

6. PATIENT PREAPARATION: NA

7. TYPE OF CONTAINER AND ADDITIVES: Plastic Container

8. REQUIRED EQUIPMENT AND REAGENTS: Aqueous Nitric acid (5-10%), Distilled Water.

9. ENVIRONMENTAL AND SAFETY CONTROLS: Universal safety precaution

10. CALIBRATION PROCEDURE (METROLOGICAL TRACEBILITY): NA.

11. PROCEDURE (STEPS) :

• Immediately after receiving the bone is cut into small pieces

• Small pieces of bone are put into the suspension of inorganic aqueous Nitric acid (5-10%)

• Solution should be able to make contact with all surfaces of a specimen

• Bone is suspended in solution for 24 hour in the Nitric acid

• Check for the completion of Decalcification

• Physically by palpating or needling the tissue

• Follow routine procedure for processing of the bone
