Preparation of Aliquots
Write sample id on ependroff cups with permanent marker pen on the rough surface of ependroff cup.
Arrange them in ascending order in the rack.
Aspirate serum from the centrifuged vacutte with the help of 200 uL pipette.
Dispense the serum into the respective sample id ependroff cup.
Recap the ependroff cup as soon as possible after dispensing serum.
Put ependroff cups with cap in refrigerator(2-8 ') if analysis is delayed.
Batch calibration in Erba-XL 640
Put 1 cup of Blank
Put 1 cup of Cal 2
Put 1 cup of Cal 3
Put 1 cup of QC 5
Put 1 cup of QC 8
Run above Cups as sample for UIBC test and take 2 OD of each.
Export data in XL sheet and calculate factor.
To Run Patient sample select NUIBC test
You will get 26 th cycle OD.
Also note down 25th cycle OD in XL sheet so you get idea of interference of RBC in sample.If 25 th cycle od is high (> 0.4), see the sample. In hemolysed sample, 25th cycle OD will be high so do not release report and write comment in Remarks as “ sample is hemolysed, send repeat sample.If sample is not hemolysed, centrifuge it again and repeat test. If sample is QNS, do not send result.ask for repeat sample.
Care to be taken while preparing ependrof cups for testing UIBC and IRON in ErbaXL
centrifuge vacuttee, seprate serum carefully in ependrof cup and take care that RBCs are not to be present in seprated serum.
if sample seems turbid, centrifuge seprated serum further and then seprate serum from this cup in fresh another cup.
if there is huge lump in graph and later goes down after R2 it may probably show contamination with RBC.
