Checking Pepsin activity
Take fibrin clot ( from PT tube/ clotted sample) .
Take 2 small fibrin clots in 2 different glass tubes.which is shown below .clot in pepsin turn into dark red colour due to formation of acid hematin (reaction between hemoglobin with HCL present in pepsin)

Add 0.5 ml di water in 1 glass tube. This is control.
Add 0.5 ml pepsin in another glass tube . This is test sample.
Gently mix for 5 minutes at room temperature.
If clot dissolve in test glass tube and not in control sample then Pepsin is active. Dissolve clot decrease in size which is shown below.
