  1. Perform sample Centrifugation
  2. Take 1 ml reagent in 1.5 ml eppendorff cup in required numbers and bring to room temprature(30 min)
  3. Take 10 microliter sample in each tube at 30 second interval. Change tip every time.
  4. incubate at room temprature for 1 hour.
  5. Select “Run test” → Glucose in Erbachem menu.
  6. When instrument asks “Aspirate water”, aspirate reagent instead. (To correct for absorbance of reagent blank)
  7. When instument ask for reagent blank, press 'No' (To prevent automatic correction of factor)
  8. take reading at 505 nm with 30 second interval.
  9. Between two sample wash cuvet with water.
  10. Take care not to exceed 30 second between reading of two samples.
  11. Enter results( Actually absorbances) in spreadsheet
  12. Evaluate factors and QC results. If factor/QC results show major deviation from past, rerun batch.
  13. If sample absorbance more than 2.0 then dilute the sample about 1:1 with DI water and rerun the sample.
  14. Calculate results=(Absorbance)X(Average Factor) in spreadsheet.
  15. instructions for determining quantitative results when a result is not within the measurement interval: Dilution of sample with DI water is done in same way as autodilution done by Erba XL-640
