====WDI OF VARIANT II HEMOGLOBIN TESTING SYSTEM==== START UP\\ -Switch on main power supply\\ -Switch on UPS\\ -Start CPU and Monitor\\ -Open CDM 5.1 program\\ -Switch on VCS(VARIANT II CHROMATOGRAPHIC SYSTEM)\\ -Switch on VSS(VARIANT II SAMPLING STATION)\\ -Click on variant II β thal Inactive mode\\ -Click on variant II β thal Inactive mode,it prompt return to active, then click yes\\ -So,the variant II β thal comes on ready mode,ok it by cliking on it\\ -Click Run\\ -In Run click worklist\\ -Wait upto green light\\ -When shows green light, click on start option\\ -Then machine is ready to run\\ -Load sample into the system racks\\ -place the following in the sample rack: ^Tube position^Adapter label^Reagent^ |1|Primer|Whole blood primer| |2|Blank|Distilled water| |3|Blank|Distilled water| |4|Cal 1|Variant A2/F calibrator| |5|Control level 1|Low control| |6|Control level 2|High control| |7 to N|Patient sample|-| |N+1|Control level 1|Low control| |N+2|Control level 2|High control| |N+3|Stop|-| -Once the run is finished ,note retention time of A2 and temperature.\\ -After making the temperature adjustment, wait 10-15 min for temperature to equilibrate and start sample run.\\ SHUT DOWN:\\ -Once the run is finished,as indicated by 'STOP'barcoded tube in rack\\ -Close CDM 5.1 program\\ -Switch turn off on both monitor and CPU\\ -Switch turn off on VCS and VSS\\ -Switch turn off on UPS\\ -Switch turn off on main power supply.\\ **MAINTENANCE**\\ *DAILY LOG CHECKLIST\\ __Prerun check:__\\ -Buffer and wash bottle levels\\ -Pressure check pump A\\ -Pressure check pump B\\ -Piston seal wash:inspect tubing for liquid(add DI water if needed)\\ -Paper supply\\ -Leaks\\ -Cartridge injection count\\ -Waste container level\\ __Postrun check:__\\ -Remove samples\\ -Wipe spills\\ __*MONTHLY MAINTAINANCE CHECKLIST__\\ 1)Clean /Decontaminate sampling fluid path:\\ -Disconnect cartridge to detector tubing from the cartridge holder\\ -Remove the analytic cartridge\\ -Clean the cartridge holder with a cotton swab and DI water\\ -Install the plastic PEEK dummy cartridge into holder\\ -Install decon tubing to the outlet of the cartridge holder\\ -Place outside of decon tubing into a breaker to collect wastw from decontamination procedure.\\ -Select the V2-DECON test from the SETUP/Test screen\\ -Click ok on the test dialog box\\ -Place five sample microvials of 5%sodium hypochlorite solution into the adapters in the first five position of a sample rack\\ -In the last five position of the rack, place five sample microvail with DI water\\ -Put the stop tube with an empty sample microvail into the second sample rack and place both sample racks on the sampling station\\ -Start worklist\\ -When the status returns to READY,remove the plastic PPEK dummy cartridge, clean the cartridge holder again with a cotton swab and DI water, then reinstall original analytic cartridge\\ -Remove decon tubing from the cartridge holder.Discard breaker waste.\\ -Reconnect cartridge to detector tubing, securing both tubing end connection\\ -Remove the racks.select the desired test from the SETUP/Test screen.\\ 2)Clean the Flow cell\\ -The flow cell cleaning kit contains a syringe, flow cell cleaning liquid and tubing assemblies:\\ -Make sure the variant II is in the READY mode.open the front panel of the VCS and disconnect the fingertight fitting connected to the outlet of the column thermomodule\\ -Remove the syringe, adapter ang union fitting from the kit.connect the adapter to the union fitting.connect a syringe filled with cleanng solution to the adapter.\\ -Using the syringe ,push about 20ml of solution through the flow cell.let stand for atleast 20 min.repeat once.\\ -Fill the syringe with DI water.using the syringe ,push about 20 ml of DI water through flow cell to rinse out the cleaning solution.Repeat once.\\ -Remove the union fitting, adapter, syringe used for cleaning and reconnect the tubing for the thermomodule outlet.\\ 3)Flush piston/Seal valve\\ 4)Clean Exterior/Interior surface/Belts\\ Exterior surface cleaning:Use a cloth or sponge dampened with water to wipe the surface.\\ Interior surface cleaning:Wipe up any fluid, using a soft disposable towel or tissue.clean the interior base plate of the system by wiping it with a soft cloth or sponge dampened with water.\\ Belts can be actuated using the cleaning selection in the MAINTAIN/Instrument screen.\\ 5)Clean Barcode Reader\\ -Remove racks from the VSS.\\ -Using a soft cotton cloth, gently wipe the barcode reader face.\\ 6)Clean Dilution chamber\\ -Power off the VSS.\\ -Remove the needle cover door\\ -Push the needle gantry back, cleaning the needle from being over the dilution chamber\\ -clean the dilution chamber using cotton swabs and DI water\\ -Replace needle cover door\\ -Power on VSS,waiting for sampler to complete intialization and flush cycle\\ -Click ok to CDM communication error box, sampler resetting box and rack resetting box, returning the system back to ready state.\\ 7)Clean and Inspect Sample Racks with a cotton swab\\ 8)Clean Sample Needle\\ -From CDM,go to the MAINTAIN/Instrument screen.\\ -Under sampler ,choose the replace needle button\\ -After the needle is repositioned ,open the automatic sampler cover of the sampling station\\ -clean the sample needle with a cloth, removing debris.\\ -Close the cover to the sample needle.Power should be restored to the unit.Choose the move needle home button. The needle will return to its home position and a message will appear asking if you wish to flush the sampler. It is recommended that you perform this flush.
