====FLUID PROCESSING==== -**PURPOSE:** To prepare the slide from received body fluid, sputum and BAL fluid -**STAFF RESPONSIBLE:** Senior technician. -**PRINCIPLE:** Sedimentation -**REAGENT:** Glacial acetic acid -**PERFORMANCE SPECIFICATION:** 2000rpm for 10 mins *Linearity: NA *Precision and accuracy: NA *Sensitivity/specificity: NA -**SAMPLE:** Body fluid, BAL, Urine,Bladder wash, CSF -**TYPE OF CONTAINER:** Wide mouth sterile container -**EQUIPMENT AND REAGENT USED:** Conventional centrifuge, cytotek centrifuge,slides, applicator, WBC diluting, specimen, chamber holer, filter paper. -**CALIBRATION PROCEDURE:** Calibration of centrifuge is done regularly. -**PROCEDURE (STEPS):** *Conventional centrifuge *Take the fluid into the test tube. *In case of Hemorrhagic fluid, add equal volume of glacial acetic acid to the fluid. *Centrifuge it at 2000 rpm for 10 min. *Remove the supernatant, mix the sediment with the help of vortex mixture, take one drop on slide, prepare the smears. -**QUALITY CONTROL PROCEDURE:** When new -**REFERENCE INTERVALS:** NA -**TAT FOR TEST:** Immediately processed -**LABORATORY INTERPRETATION:** NA -**ALERT CRITICAL VALUE:** NA -**SAFETY PRECAUTION:** Universal safety precaution (Biosafety manual). -**VALIDATION OF RESULTS TO BE DONE BY:** By authorized signatories &Assistant Technical manager. -**REVIEW, RECORDS & RECOMMENDATION:** As per ISO:15189 Guidelines Clause 4.13 & NABL 112 -**Reference:** -Theory & practice of Histological Techniques, 4th edition, 1996, John D.
