====SOP of Fluid processing==== **1.PURPOSE** To prepare the slide from received body fluid, sputum and BAL fluid **2.STAFF RESPONSIBLE** Senior technician. **3.PRINCIPLE** Sedimentation **4.REAGENT** Glacial acetic acid **5.PERFORMANCE SPECIFICATION** NA **6.SAMPLE** Body fluid, BAL, Urine,Bladder wash, CSF **7.TYPE OF CONTAINER** Wide mouth sterile container **8.EQUIPMENT AND REAGENT USED** *Conventional centrifuge, cytotek centrifuge, *slides, applicator, WBC diluting, specimen, chamber holer, filter paper **9.CALIBRATION PROCEDURE** Calibration of centrifuge is done regularly. **10. PROCEDURE (STEPS)** __==A. Conventional centrifuge==__ -Take the fluid into the test tube. -In case of Hemorrhagic fluid, add equal volume of glacial acetic acid to the fluid. -Centrifuge it at 2000 rpm for 10 min. -Remove the supernatant, mix the sediment with the help of vortex mixture, take one drop on slide, prepare the smears. **11.
