====Ketones==== ===Reference=== ==Preparation of Rothera's power== - Weight following chemical & mix thoroughly - Sodium carbonate ­­­­­­­= 20 gm - Ammonium sulfate ­­­­= 20 gm - Sodium nitroprusside ­­­= 1 gm (make fine powder of it with mortar and pastle before mixing) - ==Sample== -Urine -Serum(Blood) [[nchsls:c:biochemistry:document:online_information_resource_for_patients_and_users#Instructions_for_Patient_Collected_Samples|Special instructions for patients]] ==Procedure== -Take pinch of powder on a glass surface or a tile or a tile with well in it -Put a drop of sample on powder -Do not take sample more than what powder can absorb. ==Observation and interpretation== -If purple color observed, ketones are reported as "positive" -If no purple color observed, ketones are reported as "negative" -Read IQC (as shown below) to prevent confusion in detection of color ==Internal Quality Control== -Frequency of IQC : Whenever sample is received, Both the control (Positive & negative ) has to be run along with test. -Prepare 20 ml urine pool from OPD samples of apparently healthy individual. Divide in to two parts. -**Preparation of Positive control** -In 10 ml of pooled urine- add 10 ul acetone. -Prepare aliquotes of 200 ul and give lot number e.g KP1, KP2, KP3.... - -**Preparation of Negative control** - Take 10 ml of pooled urine - Prepare aliquotes of 200 ul and give lot number e.g KN1, KN2, KN3.... -Take powder in two wells. -In one well add negative control. -In another well add positive control. -Note observation in Ketones IQC Sheet -Upload sheet every month in LIS{{:nchsls:c:biochemistry:document:ketone_iqc_sheet.odt|Print sheet}} -The monthly QC sheet has to be uploaded in KTO calibration file. ==External Quality Control== Currently there is external quality Control done in the laboratory.
