====How to make Electrolyte pack==== {{{{:nchsls:c:biochemistry:document:electrolyte_pack_preparation_mops_hepes.ods|}}|Preparation of electrolyte pack(Erba Xl-640)}} ====Required Glassware==== - 1000 ml volumetric flask - Funnel - 2 Glass bowl - 500 microliter pipette ====Procedure==== - Put Nacl and Kcl in Glass bowl.Put Glass bowl in Hot air oven at 175'c for 1 hour.(To evaporate water present in Nacl and Kcl) - Take out Glass bowl carefully with holder or handkerchief.(To prevent injury to ourself). ===Std A=== - Weight following chemicals in given volume and mix in plastic container - 8.123 gm of Nacl - 0.298 gm of Kcl - 0.065 gm sodium azide - Make such 10 cups and preserve in 2-8 degree temperature ===During reagent making=== - Take 600ml DI water in 1000ml volumetric flask and add contains of preprepared dry chemicals fron plastic cups. - Add 36.95 ml of Lithium carbonate stock solution(1 gm/L) in volumetric flask. - Mix well and make sure that all chemicals are completely dissolved. - Make upto 1 liter with fresh DI water. - Record data in [[inventory management|inventory management section of LIS]] ===Std B=== - Weight following chemicals in given volume and mix in plastic container - 4.032 gm of Nacl - 0.596 gm of Kcl - 0.065 gm sodium azide - Make such 5 cups and preserve in 2-8 degree temperature ===During reagent making=== - Take 600ml DI water in 1000ml volumetric flask and add contains of preprepared dry chemicals fron plastic cups. - Add 14.78 ml of Lithium carbonate stock solution(1 gm/L) in volumetric flask. - Mix well and make sure that all chemicals are completely dissolved. - Make upto 1 liter with fresh DI water. - Record data in [[inventory management|inventory management section of LIS]] ===Stock lithium carbonate(1 gm/L)=== - Weight 1 gm lithium carbonate and add it into approximately 600 ml water into volumetric flask. - Make upto 1 liter with fresh DI water. ===Quality check of electrolyte reagent=== - After making electrolyte reagent , Take old and new A and B 0.5 ml in 4 ependroff cups.. - Measure electrolyte in all 4 cups. - Store data in "10.207.3.240/NCHSLS/c/001 Biochemistry/Records/009 Internal Quality Control Records/Electrolyte QC check". ====Preparation of ISE Cleaning solution(DO NOT USE THIS) ==== ==Preparation of ammonium bifluoride stock solution== -Take ammonium biflouride powder bottle from refrigerator.Open only in presence of Head of department as it is very harmful chemical. -Dont use any glass containers for making ammonium biflouride stock solution as ammonium biflouride erodes the glass. -Take 500 ml DI water in 500 ml glass volumetric flask(as we dont have plastic 500ml volumetric flask and we are using this flask for measuring DI water and rest of the procedure done in plastic beaker) and take out 21.5 ml DI water with 10 ml graduated pipette with pipetter. -Add remaining DI water of 500 ml volumetric flask in 1 liter of plastic beaker. -Add 21.5 ml 10% HCL(35% W/W, Density=1.18 , MW=36.46) in 1 liter of plastic beaker. -Add 1.425 gms ammonium bifluoride(MW=57.0) in 1 liter of plastic beaker and dissolve it by tip of 500 ul pippette. -This is stock solution of 0.05N ammonium bifluoride+0.05N Hcl. -Fill 15 ml DI water in 15 ml plastic reagent bottle with 10 ml greduated pipette with pipetter.and note the mark on that bottle.Use this mark as reference for filling other bottles with ammonium biflouride stock solution. -Fill approximately(as described above) 15 ml of this solution in 15 ml reagent plastic bottle directly from 1 liter of plastic beaker. -Make such 32-35 bottles -reagent expiry is up to 1 year. -Record data in [[inventory management|inventory management section of LIS]] ==Preparation of pepsin eppendroff== -Take 0.420 gms Pepsin(1:10000) powder in each eppendroff cups and make 30 such cups.seal with parafin film. -Put pepsin eppendroff cups and 15 ml bottles of ammonium biflouride stock solution in box and paste {{instruction for making ISE solution.odt|instruction}} on the box. ==Preparation of working ISE cleaning solution== -Mix 1 eppendroff cup of pepsin powder in 15 ml bottle of ammonium biflouride stock solution ==Checking Pepsin activity== -Take fibrin clot ( from PT tube/ clotted sample) . -Take 2 small fibrin clots in 2 different glass tubes. -Add 0.5 ml di water in 1 glass tube. This is control. -Add 0.5 ml pepsin in another glass tube .
